The latest title from the acclaimed Current Protocols series, Current Protocols Essential Laboratory Techniques, 2e provides the new researcher with the skills and understanding of the fundamental laboratory procedures necessary to run successful experiments, solve problems, and become a productive member of the modern life science laboratory. From covering the basic skills such as measurement, preparation of reagents and use of basic instrumentation to the more advanced techniques such as blotting, chromatography and real-time PCR, this book will serve as a practical reference manual for any life science researcher. Written by a combination of distinguished investigators and outstanding faculty, Current Protocols Essential Laboratory Techniques, 2e is the cornerstone on which the beginning scientist can develop the skills for a successful research career.
Electrophoresis is a straightforward but informative analytical method used in biochemistry, biology and medicine. This book combines a detailed discussion of theory and technical application with an elaborate section on troubleshooting and problem solving in electrophoresis. Therefore the book is an important guide for both students and scientists.
"Electrophoresis in Practice" ist seit mehr als zwei Jahrzehnten das Standardwerk in der Elektrophorese. Die 5. Auflage wurde sorgfältig überarbeitet und beinhaltet nun ein erweitertes Kapitel zu Mikromethoden und der chipgebundenen Elektrophorese.
It is a truism of science that the more fundamental the subject, the more universally applicable it is. Neverthelens, it is important to strike a level of "fundamentalness" appropriate to the task in hand. For example, an in-depth study of the mechanics of motor cars would tell one nothing about the dynamics of traffic. Traffic exists on a different "level" - it is dependent upon the existence of motor vehicles but the physics and mathematics of traffic can be adequately addressed by considering motor vehicles as mobile "blobs", with no consideration of how they become mobile. To start a discourse on traffic with a consideration of the mechanics of motor vehicles would thus be inappropropriate. In writing this volume, I have wrestled with the question of the appropriate level at which to address the physics underlying many of the techniques used in protein isolation. I have tried to strike a level as would be used by a mechanic (with perhaps a slight leaning towards an engineer) - i.e. a practical level, offering appropriate insight but with minimal mathematics. Some people involved in biochemical research have a minimal grounding in chemistry and physics and so I have tried to keep it as simple as possible.

Immunological Methods a compendium of basic research techniques being used in one of the largest immunology research institutes, the Basel Institute for Immunology, with particular emphasis given to new methodology. The procedures have been described by individuals judged to be highly expert in their specialties. In many instances the methods developed or adapted to unique uses by the contributors have not previously been described in detail. The book contains 34 chapters covering techniques for detection, isolation, and purification of antibodies (including dansylation, two-dimensional chromatography, isoelectric focusing, polyacrylamide gel electrophoresis, and isotachophoresis); measurement of equilibrium constants (equilibrium dialysis, filtration, and sedimentation); and isotope and fluorescent labeling and detection of cell-surface components. Techniques such as isotope laboratory maintenance; chemical modification of proteins, haptens, and solid supports, and haptenation of viable biological carriers; production of antisera against allotypes and histocompatibility antigens and production of antibody with clonai dominance; histocompatibility and MLR testing; and cell separation by haptenated gels and by velocity sedimentation of rosette-forming cells are also discussed. Other chapters cover detection of antibody-secreting and alloantigen-binding cells; immune responses in vitro and their analysis by limiting dilution; production of T-cell factors; hybridoma production by cell fusion; maintenance of cell lines and cloning in semisolid media; and the mathematical analysis of immunological data.
Microchemical Analysis of Nervous Tissue focuses on the use of microbiochemical methods in the analysis of nervous tissue, with emphasis on those related to the study of amines, amino acids, phospholipids, and proteins. Special attention is paid to the choice of biological material and the various procedures used for the isolation by dissection of defined components of the nervous system. Comprised of 10 chapters, this volume begins with an overview of microprocedures used in neurochemistry, followed by a discussion on the importance of choosing the biological material for microanalysis. The isolation of nervous tissue for analysis is then considered, with particular reference to invertebrate neurons; cell components from fresh, "fixed," freeze-dried, and frozen impregnated tissue; and discrete areas of nervous tissue. Subsequent chapters describe some instruments and glassware used in microprocedures, along with the applications of such procedures; general techniques used in microprocedures; microdetermination of phospholipids as well as amines and amino acids as dansyl derivatives; and microelectrophoresis of proteins. This book will be of interest to molecular biologists, microbiologists, physiologists, and neurochemists.
