The social, political, and cultural consequences of attempts to cheat death by freezing life. As the planet warms and the polar ice caps melt, naturally occurring cold is a resource of growing scarcity. At the same time, energy-intensive cooling technologies are widely used as a means of preservation. Technologies of cryopreservation support global food chains, seed and blood banks, reproductive medicine, and even the preservation of cores of glacial ice used to study climate change. In many cases, these practices of freezing life are an attempt to cheat death. Cryopreservation has contributed to the transformation of markets, regimes of governance and ethics, and the very relationship between life and death. In Cryopolitics, experts from anthropology, history of science, environmental humanities, and indigenous studies make clear the political and cultural consequences of extending life and deferring death by technoscientific means. The contributors examine how and why low temperatures have been harnessed to defer individual death through freezing whole human bodies; to defer nonhuman species death by freezing tissue from endangered animals; to defer racial death by preserving biospecimens from indigenous people; and to defer large-scale human death through pandemic preparedness. The cryopolitical lens, emphasizing the roles of temperature and time, provokes new and important questions about living and dying in the twenty-first century. ​This book includes papers from keynote lecture and oral presentations of Plant and Microbe Adaptations to Cold (PMAC) 2012, an international conference on winter hardiness of crop and pathogenic microbes. The PMAC has been started in 1997 in Japan as an interdisciplinary forum for scientists and extension people working in the field in plant pathology, plant physiology, microbiology, and crop breeding to increase our knowledge and improve our understanding of overwintering of crops, forages and grasses and solve the problems associated with losses due to freezing and heavy snow cover. Successive meetings have been held in Iceland (2000), Canada (2003), Italy (2006), and Norway (2009). PMAC2012 will be a special meeting with a focus on global climate change, food security and agriculture sustainability and the whole program will be arranged to reflect this theme. In the late 18th century explorers and scientists started venturing into the Arctic in a heroic and sometimes deadly effort to understand and unveil the secrets of the unforgiving and mysterious polar region of the high north. Despite that the Arctic was already populated mattered less for the first wave of polar researchers and explorations who nevertheless, brought back valuable knowledge. Today the focus in Arctic science and discourse has changed to one which includes the peoples and societies, and their interaction with the world beyond. The image of a static Arctic - heralded first by explorers - prevailed for a long time, but today the eyes of the World see the Arctic very differently. Few, if any, other places on Earth are currently experiencing the kind of dramatic change witnessed in the Arctic. According to model forecasts, these changes are likely to have profound implications on biophysical and human systems, and will accelerate in the decades to come. “The New Arctic” highlights how, and in what parts, the natural and political system is being transformed. The book reproduces 55 of more than 300 articles written by the author, representing milestones in methods development of single-particle cryo-EM as well as important results obtained by this technique in the study of biological macromolecules and their interactions. Importantly, neither symmetries nor ordered arrangements (as in two-dimensional crystals, helical assemblies, icosahedral viruses) are required. Although the biological applications are mainly in the area of ribosome structure and function, the elucidation of membrane channel structures and their activation and gating mechanisms are represented, as well. The book is introduced by a commentary that explains the original development of concepts, describes the contributions of the author's colleagues and students, and shows how challenges were overcome as the technique matured. Along the way, the ribosome served as an example for a macromolecule with intricate structure and conformational dynamics that pose challenges for three-dimensional visualization. Toward the end of the book -- bringing us to the present time -- molecular structures with near-atomic resolution are presented, and a novel type of computational analysis, manifold embedding, is introduced. Single-particle cryo-EM is currently revolutionizing structural biology, presenting a powerful alternative to X-ray crystallography as a means to solve the structure of biological macromolecules. The book presents in one place a number of articles containing key advances in mathematical and computational methods leading up to the present time. Secondly, the development of the technique over the years is reflected by ever-expanding discoveries in the field of ribosome structure and function.
